Does anyone know if I can run a 40 Gb hard drive with my Asus P5a-b motherboard? I was thinking I may have to update the BIOS to do so. Anyone actually have to do this?

Support IDE HDD larger than 32G Bytes.

so yes if your not running bios 1010 then you need to upgrade to it for 40gig support
