I have 4 dimm slots for RAM on my mobo. Now according to the manual I should be able to place my stick of RAM in which ever slot I choose and it should work fine. Right? In my case WRONG! I tried this (last ditch effort with all the problems I've been having) and my comp will not boot up unless it is in the first slot.

Anyone else had this problem?
Does this mean that something has been to my RAM to necessitate it being in that specific slot?

a lot of boards are like that. it's the board not the ram. just put it in the first slot. many times, the largest module has to be in the first slot.

Yes, some manuals say that you can put 1 stick of RAM in any slot, but sometimes that won't work even though the manual says it will work.

My board won't boot unless 1 stick is in slot 1. If I have a 2nd stick then it can go into any other slot I want.

To answer your question regarding RAM placement, there are many things that affect if or not the memory you installed would work. Several factors have to be taken into consideration, mobo chipset, design and BIOS affect how the memory is recognized and how fast it makes the whole system work! If you did your research correctly, there were articles where DDR333/PC2700 memory based mobos outperformed the DDR400/PC3200 memory based mobos! You could check out the articles at the following websites: Toms Hardware Guide, Firing Squad and Anandtech.

They've featured articles that compared the difference in performances between the above mentioned mobos! It should be able to shed some light to your question.:)
