Is it necessary or possible or "practical"
to alter the settings/BIOS on a motherboard so
that it will recognize and use larger SDRAM DIMMS?

Specificly, AMI BIOS 063100, Trigem NAPOLI 1.0
motherboard. (emachine 500MHz)

Supposedly it was designed to two 128MB DIMMs.
I currently have a 512MB and a 256MB installed
but Windows Perf Meter only sees 512MB of memory.
Other programs don't appear to see all the
memory either.

Any ideas and insight appreciated.

Of course it is possible to edit and improve on your system BIOS. If you spend the time, money, effort, buy equipment, etc. to do it like the manufacturers do.

Depends whether the MB was designed for that much RAM. If it was, all you have to do is enter BIOS then save settings and exit and it should recognize the new RAM.

The maximum amount of RAM -- both in total and per DIMM slot -- is determined by the chipset, not the BIOS, and upgrading the BIOS will not change that.
